We are looking for a highly motivated PhD candidate interested in preparative bioorganic chemistry and interdisciplinary research. The project involves the development of ligation methods for chemical protein synthesis and scientific services in research within the DFG-funded project “Catalytic and proximity-accelerated native chemical ligation”

Requirements: Completed university degree (master or equivalent) in the field of (bio)organic chemistry with preferably very good grades, theoretical and practical experience in solid-phase peptide synthesis is desirable; openness to interdisciplinary research projects in the field of natural sciences and high motivation for independent scientific work; very good communication and teamwork skills; very good written and spoken English skills.

PhD position in chemical and biological peptide science

A position for doctoral candidate  isavailable fully funded by the Austrian Science Funds project P36736-B. The project management seeks for a highly motivated graduate student who wants to join the research team of Dr. Roland Hellinger (Group leader) at the Center for Physiology and Pharmacology of the Medical University of Vienna.

The research area of the PhD thesis. Today’s drug development research is increasingly interested in bioactive peptides as a potential starting point for innovative therapies. Cyclic cystine rich knot peptides are inhibitors of the prolyl oligopeptidase (POP) and have modulatory activity on the protein network. The active-site directed inhibitors of POP were shown to reduce alpha-synuclein oligomerization in vitro as well as in in vivo models. However, a modulation of POP activity, by a stabilization of conformational states, and thus its protein network interactions, was identified as the main factor for the clearance of toxic particles and improved neuronal autophagy. Within the frame of this project a range of models will be applied to study peptidic modulators and a POP dependent mechanism, which affects alpha-synuclein aggregation and autophagy.

Requirements.

The candidate will perform a PhD thesis in the field of chemical and biological peptide science. Natural or synthetic peptides will be prepared for bioactivity testing with various in vitro assays using recombinantly expressed proteins or cellular reporter assays. The candidate will utilize techniques, e.g. mass spectrometry, flow cytometry, reporter assay, qPCR and molecular cloning, solid phase peptide synthesis, liquid chromatography, protein chemistry and other molecular biology methods.

The successful candidate should hold a Master/Diploma’s degree in biology, molecular biology, biotechnology, biomedicine or related subjects. Ideally, the applicant has experience in one or several of these key technologies: protein chemistry, mass spectrometry and chromatography or peptide chemistry. Laboratory working experience is of ultimate need to rapidly implement the planned research activities. The applicant will work on a multi-disciplinary project, embedded in a scientific research group and an international research team.  

Additional Information

The research group is located at a top-level research institution (Medical University of Vienna). The institution guarantees a high-level scientific environment in which the PhD candidate will develop to an independent scientist. The candidate will work as scientific staff, be enrolled in a thematic PhD program and participates in regular research seminars, journal clubs and other training opportunities. We offer remuneration in accordance with collective bargaining agreement: §48 VwGr. B1 Grundstufe (praedoc) (max. 30h/week) with relevant work experience determining the assignment to a particular salary grade.

We are offering a three-year PhD fellowship in Sustainable Peptide Chemistry commencing 1st April 2024 or as soon as possible thereafter.

Project description Our laboratory has longstanding experience in the synthesis of biologically active peptides and peptide analogues, developing potent and selective peptide-based ligands against a series of different targets. However, one of the major challenges in peptide and protein science is the consumption of reagents needed for the preparation of chemically modified candidates that are not amenable to expression. In this project we will seek to take advantage of electrochemical methods for the modification of peptides and proteins, to develop novel avenues for large-scale introduction of non-canonical chemical modifications into biologically active peptides or proteins without the consumption of harmful reagents and organic solvents.

Duration 3 years as a PhD student.

Our group and research With a foundation in chemical synthesis, the Olsen Lab seeks to explore the function of small molecules, peptides, peptidomimetics, and proteins in biological systems. Thus, working in the interdisciplinary interface between chemistry and biology. We are exploring naturally occurring macrocyclic peptides and analogues thereof for development of class- or isoform-selective HDAC inhibitors and quorum sensing modulators. We are also investigating protein post-translational modifications in the context of epigenetics and metabolism, as well as the substrate-specificity of the human histone deacetylase enzymes. The laboratory is currently comprised of about ten M.Sc. students, Ph.D. students, and postdoctoral fellows of various nationalities, with backgrounds in chemistry, biochemistry, and engineering. We take pride in working as a team and having a balanced gender composition.

Our department Department of Drug Design and Pharmacology is committed to research-based teaching and interdisciplinary research that supports the development and understanding of chemical and pharmacological properties of drugs and drug targets. We work as part of a dynamic academic community of research, education and innovation within the Pharmaceutical Sciences bridging Health Sciences and Life Sciences.

Job description Your key tasks as a PhD student at the Faculty of Health and Medical Sciences are:

  • Carrying through an independent research project under supervision
  • Completing PhD courses or other equivalent education corresponding to approximately 30 ECTS points

Salary, pension, and terms of employment are in accordance with the agreement between the Ministry of Finance and The Danish Confederation of Professional Associations on Academics in the State. Depending on seniority, the monthly salary starts at approximately 28,993 DKK/roughly 3,890 EUR (October 2023 level) plus pension.

John obtained his PhD in Biochemistry in 1980 at Monash University. He then received a Nuffield Foundation Fellowship to Cambridge, UK, at the MRC Laboratory of Molecular Biology, working with RC Sheppard on modern methods of chemical peptide synthesis. In 1983 he returned to Australia to the Howard Florey Institute (now The Florey), University of Melbourne, where he has long headed the Peptide and Protein Chemistry Group. His interests are in chemical peptide synthesis of complex, functionalised peptides, including antimicrobial peptides. Professor Wade is a past NHMRC of Australia Principal Research Fellow and a Fellow of both the Royal Australian Chemical Institute and the Royal Society of Chemistry.
